Central Thyrotrophin-Releasing Hormone Participates in Obesity-Induced Hypertension.

Diencephalic thyrotropin-releasing hormone (TRH) is involved in central cardiovascular regulation and also controls energy expenditure. Recently we demonstrated that TRH interacts with leptin and may be important in the pressor effect produced by leptin. Here, to study the role of TRH in obesity-induced hypertension (OIH), we used a simple model of OHI produced by a high-fat diet (40% w/w) during 45 days in 36 male Wistar rats in comparison with 6 controls (C) which were under a standard diet. Results are expressed as mean -SD. Body weight and systolic arterial blood pressure (ABP) were 330-44 vs 268+/-31 g (p<0.04) and 161 +/-10 vs 137+/-14 mmHg (p<0.04), respectively. Plasma leptin (ElA) correlated with peritoneal adiposo tissue (ir: 0.791, n=30, p<0.001). With the aim of studying the participation of TRH (RIA) in this OIH model, we compared with C, 18 OIH animals divided in 3 groups treated with: V, vehicle AS, an antisense oligonucleotide against the preTRH and INV, the inverted AS sequence. Table shows that the AS significantly decreased TRH content at 48 hs after treatment along with the ABP at 24 or 48 hours. In contrast, vehicle and INV produced no changes. These AS-induced n ABP were not due to an alteration in the thyroid status since plasma T3 and T4 levels remained unaltered (data not shown).
