EFFECT OF BETA PINENE, ISOLATED FROM TILIA CORDATA MILL. FLOWERS, ON NORMAL AND TUMORAL LYMPHOCYTES PROLIFERATION

Resumen:
Tilia cordata is a plant used in popular medicine for anxiety and as immunostimulant. In a previous work, we demonstrated that, a dichloromethanic extract obtained fromT.cordata flowers presented a selective antiproliferative action on a murine lymphoma cell line (BW 5147). The terpenes limonene and beta pinene (bp) were identify by gas chromatography mass. The aim of the present work was: a) to analize the effect of isolated b p on BW 5147 cells and on normal murine lymphocytes proliferatio, through tritiated tymidine uptake, b) to determine cytostatic or cytotoxic effects by tripan blue exclusion method, d) to investigate the induction of apoptosis by nuclear Hoescht dye assay and e) to analyze  the effect on total nitrites levels by the Griess method. Results (media ± SEM): tumoral cells (CI50 ug/ml, 24 hs: 79.4 ± 5.0; 48 hs: 15 ± 1.2; 72 hs; 6±0.5). Normal cells: Sitmulation index (SI) (b p 0.01 ug/ml: 1.4 ± 0.08; b p 0.1: 1.65±0.07 ug/ml; b p 1 ug/ml: 3.4 ± 0.3; b p 10 ug/ml 3.2 ± 0.2; b p 100 ug/ml: 2.4 ±0.18: b p 1000 ug/ml: 2.3 ±0.2). Total nitrites 24 hs SI (b p 0.01 ug/ml: 2.2 ± 0.2; b p 0.1: 2.07±0.18 ug/ml; b p 1 ug/ml: 1.06 ± 0.08; b p 10 ug/ml 0.7 ± 0.05; b p 100 ug/ml: 0.70 ±0.06). b p presented cytostatic and cytotoxic effect, these effects were related with apoptosis and  the level of total nitrites.
