Resumen:
The occurrence of high metal amounts in plants, principally edible varieties, is of considerable importance, since they might constitute a possible toxicological hazard. Metals, when concentrated in living organisms are often considered toxins toward mammals, particularly humans. Many studies have dealt with the determination of total elemental content of mushrooms, in particular the edible ones. However, information about elemental speciation in edible mushrooms is scarce.In this work, fractionation of soluble species of Ag, Ba, Bi, Br, Cd, Cu, Fe, Hg, I, P, Pb, Ni, and Zn found in edible mushrooms was done by on-line coupling of size-exclusion chromatography (SEC) and ICP-MS. Different extraction procedures were developed with NaOH, HCl, and double distilled water. The separation was performed on a Superdex 75 column (300 x 10 mm) with 0.010 mol l-1 CAPS buffer at pH 10 as the mobile phase. All the variables involved into the SEC separation, ICP-MS detection, and extraction procedures were studied and their effects evaluated.For Se, the mushroom contained 19 ìg/g total element. Proteinase K enzymatic extraction of the mushroom was utilized to hydrolyze the peptide bond of each amino acid in the protein sequence. The filtered hydrolyzate was separated by an Agilent 1100 HPLC utilizing reverse phase ion-pairing chromatography with a hexanesulfonic acid/citric acid buffer at pH 3.5. Various Se species will be reported.
