In muscle cells, insulin and contraction promote glucose influx through the GLUT4 transporter. While ELISA-likeassays and immunofluorescence are the most used methods to quantify GLUT4 at the cell surface, flow cytometry hasnot been widely used for this purpose. Flow cytometry is a highly versatile technique that is widely used for a varietyof purposes. It can analyze many events per second and measure multiple parameters simultaneously. Unlike traditionalmethods, flow cytometry provides more robust and objective results in less time. In this study, we investigated anddeveloped a flow cytometry protocol to quantify GLUT4 at the plasma membrane of muscle cells, which was tested inseveral experimental conditions. The results show that flow cytometry is a simple, robust and reliable technique thatcan be used to quantify GLUT4 changes at the muscle cell surface.
