A rat model of paternal diabetes to study the programming of metabolic disorders in the offspring

Diabetes Mellitus is one of the most concerning chronic diseases worldwide. Studies about theprogramming of the offspring health from diabetic fathers are starting to appear. In the context ofa project that aims to study this issue, this work is a first approach in the knowledge of therelationship between paternal diabetes and their offspring's metabolic health.Methods: Male diabetic rats (obtained by Streptozotocin injection, 90 mg/kg) and male controlrats (injection with citrate buffer as vehicle) were mated with healthy female rats (n=8). Glucosewas measured in males in blood from the tail vein by reactive strips. After pregnancy confirmation,males were weighed and euthanized, and the testis, kidneys, liver and heart were weighed.Sperm was collected by perforation of cauda epididymis to count spermatozoa and measuremotility. On day 21 of gestation, pregnant rats were euthanized and the fetus were sexed. Theplacenta and the fetal organs (liver, heart and kidneys) were weighed.Results: The glycemia in diabetic males (519 ± 88.9) was higher than in controls (105 ± 9.7)P
