Supercritical CO2 pretreatment and subcritical water extraction of sulfated flavonoids from Flaveria bidentis leaves

Flaveria bidentis is a plant species that contains sulfated flavonoids (SFs) with therapeutic potential, besides being an invasive plant. The extraction of these active principles would add value to the biomass generated during its weeding. This work reports the optimization of the pretreatment of F. bidentis leaves with supercritical CO2 (P-SC-CO2), with the subsequent extraction with subcritical water (SWE) of these SFs. The combined effect of different levels of three P-SC-CO2 factors: time, pressure and depressurization rate, were investigated using response surface methodology involving a two-replicate 23 factorial experimental design. Optimal extraction conditions were the highest levels tested for each variable under study: Time = 30 min, Pressure = 30 MPa and Depressurization rate = 2 kg CO2/h. Under these conditions, 72.45 ± 0.99 mg of total SFs/100 g of leaves were recovered. Likewise, we were able to demonstrate that P-SC-CO2 allowed practically doubling the extraction yield of total SFs with respect to the extractive method in the absence of pretreatment, and we were able to visualize the structural damage caused by P-SC-CO2 on the plant matrix.
