Bacterial Superantigens (SAgs) are enterotoxins that interact simultaneously and in a non-classical way with the TCR Vb chain andthe MHC-II. As this interaction occurs, it induces an exacerbatedT cell proliferation and a massive cytokine release producing ToxicShock Syndrome, among other affections. The present study aimedto analyze the capacity of Staphylococcal enterotoxin N (SEN),which belongs to the egc operon, to activate the immune system byevaluating its interaction with PBMCs from healthy donors. For thispurpose, SEN was cloned in a prokaryotic plasmid, produced as a recombinant protein in E. coli and purified by Ni++/NTA column followed by Size Exclusion Chromatography. Further, SEN was identified by Mass Spectrometry. PBMCs from donors non-exposed toSAgs were incubated with different concentrations of SEN to evaluate proliferation in presence of [methyl-3H] thymidine. Supernatantswere collected for quantitative analysis of IL-6, IL-12, IL-10, TNFaand IFNg using ELISA.
