Resumen:
As mechanisms of proliferation and intracellular pHi are associatedis possible that a chronic alteration of the pH modifies cell growth. We previously showed in renal cells that aquaporin 2 water channel(AQP2) expression prevents inhibition of cell growth in acidic media.Ca2+ channel TRPV4 has been proposed as one possible acidsensor. Our previous work showed that TRPV4 and AQP2 interact. So, the aim of this study was to investigate whether renal cell growthin acidic media was dependent on TRPV4. To investigate this weused two renal cortical collecting duct cell lines: WT-RCCD1, notexpressing AQPs, and AQP2-RCCD1 expressing constitutive AQP2in the apical membrane. Cell Growth studies (CG) were performedin control and acid media in presence of vehicle or the TRPV4 inhibitorRutenium Red (RR). At physiological pH inhibition of TRPV4decreased both lines cell growth about 20%. .After 24h exposure toan acidic media (ΔpH = 0.4) only WT cells decreased its growth andthis response was not modified by RR (%CG Acid = -32±7 vs Acid+ RR= -37±9, ns n=18). On the other hand, while acid media didnot alter AQP2 expressing growth acid + RR significantly decreasedcell growth (%CG Acid =-3±4 vs Acid + RR= -51±15, p
