The study of the tumor microenvironment is important to understandthe biology of cancer. It has been known that the adipose tissue ofbreast cancer obese patient has higher TNF expression than thelean adipose tissue of breast cancer patient. Therefore, we havestudied the effect of TNF on the lipid metabolism of murine mammaryadipose tissue. In this model, TNF decreased adipocyte sizeand total lipid content, and increased glycerol secretion. With theaim of getting insights into the molecular pathways by which TNF isinvolved in the lipid metabolism of adipocytes, we used an in vitromodel of 3T3-L1 cells differentiated into adipocytes. Hence, 3T3-L1cells were stimulated with a differentiation cocktail and, after oneweek, the medium was changed for medium with or without TNF 20ng/ml for 72 h.
