Resumen:
Background:There is a correlation between hypertension and environmental pollution.Hexachlorobonezene (HCB) is a dioxin-like environmental pollutant, endocrine disruptor and regulator of thyroid hormone (HT) homeostasis. We previously demonstrated that it increases blood pressure (BP) in rats and alters its vascular morphology and function, and that TGF-β1, eNOs and REα are involved.Likewise, the effect of the toxin on the vasculature is dependent on the endothelium.Aims:To deepen the study of the mechanism of action of HCB on the vascular system.Methods:In vivo (HCB 500mg/kg by gavage 3 days a week for 30 days): a-levels of mRNA L-type calcium channels by RT-PCR. In vitro in Ea-hy926 cells (HCB 0,05; 0,5 and 5 uM for 24h): b-the expression of TGF-B1, eNOS, and REα were evaluated by Western Blot. c-the levels of mRNA TGF-B1 were analyzed by semiquantitative RT-PCR. d-nitrites levels were measured using Griess reagent and H2S by colorimetry. e-tubulogenesis: formation and of tubules in Matrigel were also analyzed.
