Natural antioxidants effect on monocyte THP-1 interleukine 1β and nitrite production

Antioxidants generally have anti-inflammatory effects. We evaluated antioxidant N-acetylcysteine (NAC) and three different sources of natural antioxidants used as infusions (Peumus boldus - boldo, Baccharis articulata - carqueja and Verbena bonariensis – verbena) on monocytes THP-1. We determined pro-inflammatory interleukin-1 (IL-1β) secretion and monocyte activation by nitrite (Nit) production. We used 5 mM NAC and natural antioxidants obtained by incubated horticultural leaves in PBS at 37°C for one hour. Antioxidant capacity was determined by DPPH percentage inhibition: 96.12 + 0.64 (NAC), 52.32 + 1.93 (Carqueja), 52.11 + 1.22 (Boldo), 48.37 + 1.46 (Verbena). After 24h treatment, carqueja increased IL-1β and Nit levels comparing to control untreated cells (12.14 + 1.61 pg/mL (carqueja) vs 8.71 + 0.35 pg/mL (control), p < 0.01) and 48.9 + 0.70 M (carqueja) vs 33.3 + 2.4 M (control), p < 0.01.), respectively. Boldo increased Nit level 57% comparing to control, but it has no effects on IL-1β. Verbena increased 23% IL-1level comparing to control, without any effect on Nit. NAC has no significant effect on IL1 or Nit production. The composition of these extracts is important to consider about their possible effects. All of the samples have high antioxidant activity; nevertheless, carqueja could be pro-inflammatory.
