Rat bone resoption (BR) measurements by urinary deoxypyridinoline (Dpd) determination. Comparison between two different comercial kits

The
knowledge of BR is very important for the treatment of bone
pathologies and its evaluation. There are different methods that
measure BR: histomorphometry, radioisotopes and bone markers, such as
Dpd, CTX and osteocalcin. Bone markers are practical but very
expensive and not always the assumptions made are correct. BR and
formation are coupled in a variety of situations, therefore bone
remodeling could be measured with one bone marker. Dpd appears as the
most suitable technique: is measured in urine, has a lower cost than
the others and two kits are available: ELISA and RIA. The former is
overpriced but requires more afordable equipment and less skillful
staff.RIA requires authorized laboratories and personnel but they are
less expensive. The aim of this study was to compare two kits of
urinary Dpd determination: ELISA and RIA, and to validate it in
biological models with modified bone remodeling. Both kits were used
to measured samples of rats from different ages, with increased BR
(by ovariectomy, by hypocalcic diet (OVX+H) and by partial
nephrectomy) and reduced BR (by treatment with zoledronate and by
parathyroidectomy (PX)). Bone remodeling was confirmed by fluoride
bone uptake and static histomorphometry.Dpd measured values
(nmol/mmol creatinine) by RIA were as expected for each model. A
significant linear regression was found in ELISA respect to RIA:
r2=0.60, slope=0.59±0.06, p<0.005. Values obtained by ELISA of
OVX+H and PX rats were not as expected. Conclusions: Values obtained
by RIA were consistent with the biological models. ELISA values, in
OVX+H and PX rats, were not as expected. ELISA could be
underestimating Dpd measurements. RIA enables greater diferences
between all the groups and is less expensive. These results indicate
that it would not be advisable to compare values obtained by this two
methods.
