Intracellular antioxidant activity and intestinal absorption of amaranth peptides released by gastrointestinal digestion in Caco-2 TC7 cells

The objective of this work was to analyse the capacity of amaranth peptides generated by gastrointestinal digestion to exert intracellular antioxidant activity and its potential modification and/or passage through the intestinal cells. A simulated gastrointestinal digested from protein isolate (APId) and its fractions (gel filtration chromatography) were analysed. The effect of a pre-treatment with amaranth samples of Caco-2 TC7 cells induced by 500 µM H2O2 was measured trough different biomarkers. All samples led to a partial or complete decrease of ROS content and different behaviours regarding the enzymatic antioxidant activities (superoxide dismutase and glutathione peroxidase) and the reduced glutathione content (increase, decrease, no effect). Fractions presented also different behaviours respect to the modification by and/or passage across Caco-2 TC7 monolayers suggesting potential actions at local intestinal and/or systemic sites. These results provide new scientific evidence that the amaranth protein isolate is a source of antioxidant bioactive peptides showing by the first time antioxidant activity at the intracellular level.
