UBA. e-Mail: dlombard@fvet.uba.ar   CFM-27 is a tumoral line obtained and characterized in our laboratory. The tumor is maintained through isogeneic transplants in Balb/cF3h mice. After a 7 day’s latency, it develops a tumor in the inoculation site, described as a poorly differentiated small cells sarcoma. Cell characterization was done with E and N-cadherines, Citoqueratine P8 and Pancitoqueratine markers. The hormone dependence was determined using anti-Progesterone receptor and anti-P29 protein monoclonal antibodies. The histopathologycal pattern is preserved among consecutive passages, which makes it a very useful tumoral model to analyze different biological aspects of hormone-dependant neoplasias. In vivo, GnRH agonists (GnRH-a) reduce levels of steroids and GnRH, simulating a chemical castration. In hormone-dependant neoplasias, GnRH-a decrease cell proliferation and increase apoptosis. Effects possibly used for therapy. According to our model characterization, the aim of this work is to analyze Buserelin Acetate (BA) regulation of tumor biological behavior. We worked with 41 animals, five weeks old, transplanted with the tumor. They were classified in three groups: two experimental groups (EG I and EG II) and one control group (CG). Treatment started when tumors reached a volume of 2.5 cm3. The dose used was 20µg/kg per day of BA for EGs, while CG received physiological solution. The treatment plan for EG I consisted in one dose per day during 7 days, no doses during the following 7 days when finally were sacrificed. Treatment for EG II, consisted in 13 doses, one per day, sacrificing them the next day. The variables assayed were animal weight, tumoral volume, histopathology, mitosis and apoptosis levels. Results show an important weight lose during days 9 to 15 for EG I which is accord to the critical mass increase of the tumor (max vol: 22.33 cm3). In EG II, the animal weight presents no variations along the experiment time, although we notice the tumoral volume increase during days 7 to 13 (max vol: 19.79 cm3). Differences between EGs and CG are no significant. Mitosis and apoptosis index will also be analyzed. With these results we could only appreciate that BA has no direct effect on biological behavior of this model.  We expect to see differences on apoptosis and mitosis levels.
