UBA. e-Mail: gangiono@fvet.uba.ar   The cellular adhesion is necessary for the organization of the CG. The Ca++ fulfils an important role stabilizing cellular unions, for example in the mediated cadherin cell contact. Alterations at this level generate changes cellular that possibly lead to apoptosis. The objective of the work was to establish in vitro the relation between the loss of the contacts dependent calcium with the integrity and the apoptosis of the CG. The study model was a cellular line of bovine CG which were treated with EGTA, with the purpose of to obtain the disruptión of the homophilic unions Ca++ dependent mediated by E-cadherin. Cellular viability (CV) was determined by exclusion with Tripan blue stain and index of apoptósis (IA) by stain of the chromatin with DAPI and TUNEL. The treatments were made with EGTA 100 nM, 9 nM and without EGTA. Parallelly the role of the depletion of serum was determined making the same study in cultures with and without bovine foetal serum (SFB) to 5%. The data were statistically analyzed by a 2x3 factorial design. Although the treatments generate changes evident of apoptósis, in the different treatments with EGTA there are statistically no significant differences between the percentage of VC average and the IA determined by DAPI. The tests by TUNEL validate the changes of apoptosis demonstrated with DAPI. In the treatments with and without SFB significant differences (p<0.051) between CV average were seen, like also for the IA (p<0.054). Since the factor serum must be considered in the study of the apoptosis, also the effect of the EGTA to different concentrations from SFB has been analyzed.
