CYTOGENETIC AND CYTOMOLECULAR TIME-DELAYED DAMAGE INDUCED IN HUMAN FIBROBLASTS BY LOW DOSES OF X-RAYS

During the past decade evidence on the non-targeted effects of ionizing radiation has been accumulated. In this study, we analysed time delayed DNA damage induced by ionizing radiation in MRC-5 Human fibroblasts cultured in vitro. We irradiated MRC-5 cells with 10 and 50 miligrays (mGy) and analysed immediately after irradiation, and at every 6 passages thereafter up to 12 passages post-irradiation by means of comet assay, micronucleus analysis and ﻻ-H2AX foci formation. Cytogenetic and cytomolecular damage were observed at all sample points analysed when compared irradiated to control cells. There were no significant differences between damage observed at each passage for all treatments. Our results let us suggest that delayed damage could be induced even by very low doses of ionizing radiation like those used in this study. Comet assay, micronucleus analysis and ﻻ-H2AX focus analysis were considered useful techniques to detect DNA damage and our results could suggest that they are also sensitive to study time delayed events like occurs in genomic instability.
