Introduction. The molecular mechanisms driving the distinct characteristic of fibroblastlikesynoviocytes (FLS) in spondyloarthritis (SpA) remain unknown. We have reported thatsynovial fluid (SF) of SpA patients induces interleukin (IL)-6 secretion by FLS.Furthermore, a proteomic study of SF of SpA showed the presence of the alarminS100A8.The objective of the present work was to study the effect of the synovial S100A8on the IL-6 production by FLS. The expression of alpha-smooth-muscle-actin (a-SMA), amiofibroblast marker, was also studied.Methods. The SF of SpA patients were pooled (n=9). Human FLS of the SW982 cell linewere stimulated with the SF pool in absence or presence of anti-S100A8/A9 antibodies.Different concentration of the S100A8 (1-10 ug/ml) were also assayed. After 24-96 h, theIL-6 levels in the supernatants were measured by enzyme-linked immunosorbent assay(ELISA). Moreover, FLS were stained intracellularly for a-SMA and analyzed by flowcytometry. The data were analyzed by the two-way analysis of variance (ANOVA)followed by Bonferroni posttest. A p value < 0.05 was considered statistically significant.Results.
