Molecular Characterization of Canine Parvovirus strains in Argentina: Detection of the pathogenic variant CPV2 in vaccinated dogs

PCR was used to detect canine parvovirus (CPV) DNA in 38 rectal swabs from Argentine domestic dogs with symptoms compatible with parvovirus disease.  Twenty seven out of 38 samples analyzed were CPV positive.  PCR amplification with sequence-specific primers allowed the identification of nine samples as CPV2a and 18 samples as CPV2b, whereas the CPV2 strain was not detected in any of the samples.  In addition, sequence analysis revealed a variant with a mutation at amino acid 426 of the VP2 gene (Asp426Glu) in 14 out of 18 of the samples initially identified as CPV2b.  This observation clearly indicates the presence of the recently described CPV2c variant in the Argentine domestic canine population.  Moreover, a retrospective analysis performed in 11 samples obtained during the years 2002-2003, showed the presence of this viral variant in one of them.  Our results identified three different pathogenic CPV variants currently circulating in the Argentine domestic dog population, with CPV2c being the predominant variant affecting vaccinated and unvaccinated dogs since the year 2007
