EX VIVO SKIN PENETRATION AND IN VIVO TOXICITY OF NANOBERRIESFOR TOPICAL DELIVERY OF ANTIOXIDANTS Purpose: To evaluate the skin penetration ex vivo of novel Nanoberries (NB), ultradeformableliposomes for skin delivery of blueberry extract rich in polyphenols, and to assess their in vivo nanotoxicity.Materials and Methods: NB were prepared by resuspension in a blueberry ethanolic extract solutionof a lipid film (soy phosphatidylcholine and sodium cholate as border activator) followed by sizeand lamellarity reduction by sonication. Human skin explants were obtained from surgery discardsand NB with double fluorescent labeling were non-occlusively applied on a SaarbrückenPenetration Model device. Blueberry extract was quantified in the stratum corneum (SC) after tapestripping. Skin slices were studied by confocal microscopy. Lethal dose and swimming activitywere determined in Zebrafish (Danio rerio) larvae model. Results: NB were about 100 nm in diameter, with low polydispersity. Skin penetration profilesshowed that the extract reached deeper layers when applied into NB in comparison to conventionalliposomes or in its free form. Confocal microscopy images are in concordance with penetration profiles. NB were non toxic at the concentration used for skin penetration. Conclusion: NB effectively entered the skin through the SC, with low toxicity. We are presentlyconducting photoprotection studies in vitro in order to determine the ability of the NB to lower thephototoxicity triggered by exposure to UV rays. NB could become a potential topical treatmentagainst ageing and damage of the skin.Keywords: liposomes, skin, antioxidants, zebrafish
