The hypothesis whether estrone (E1) could exhibit a direct action at uterus and whiteadipose tissue (WAT), under obesity was tested. The results revealed that, in obeserats, physiological concentrations of E1 rapidly increased nitric oxide (NO) synthesis,and reduced reactive oxygen species (ROS) production. The anti-oxidative action ofE1 was sustained under stress conditions provoked by an inflammatory environment orthe presence of high glucose levels. On uterine tissue, the mechanism of actiondisplayed by the steroid involves estrogen receptor (ER) participation and, depends onthe regulation of NO-ROS balance, since when NO synthase (NOS) activity wassuppressed; the reduction in ROS synthesis elicited by E1 was blunted. On WATexplants, E1 reduced ROS and thiobarbituric acid reactive substances (TBARS)production, as well as diminished leptin release. In summary, the data presentedprovide evidence that, in uterus and WAT, E1 counteracts inflammatory and oxidativestress induced by obesity.
