Effect of oral intake of Bisfenol-A on reproductive parameters of long evans rats and metabolic development of their offspring

ABSTRACT To investigate the influence of Bisphenol A (BPA) given in a soya-free rat chow on pre-pubertal development, 46 Long Evans 2 months-old female rats were divided into 3 experimental groups and fed 3 different chows: (1) control; (2) BPA 0.61 g/Kg chow (12 mg/Kg rat/day); (3) BPA 2.5 g/Kg chow (50 mg/Kg rat/day) for two and a half months. They were later mated and their offspring studied. Percentage of pregnancy and dams’ weight gain was recorded. Eyes opening was checked daily as well as vaginal opening and first estrous occurrence. At 14 days of age 6 male pups of each group were sacrificed and testis were excised, weighed and processed for testosterone determination. Pups were weaned at 22 days of age and continued into 3 experimental groups according to their mothers’ diet. At 12 weeks of age animals were sacrificed by decapitation. Spleen, kidney, pituitary gland, uterus, periuterine fat, and ovaries were collected. Plasma was also obtained for the determination of Growth Hormone (GH) and Luteinizing Hormone (LH). RESULTS: the % of pregnancy and dams’ weight gain was significantly decreased in groups 2 and 3 (p< 0.05 and p
