Reactive Oxygen Species (ROS) production and antioxidant systems in hepatocytes cell cultures from partial hepatectomised (PH) rats

Resumen:
Male Wistar rats (370-410 g ) were subjected to PH (65%) or Sh (n=5 each group). 1 hour post-surgery the isolated hepatocytes were obtained by collagenase perfusion method, and they were cultured for 1, 3, 18 and 24 hs. Results: ROS production: Sh(1h)=557±100, PH(1h)=602±65; Sh(3hs)=532±20, PH (3hs)=568±45; Sh(18hs)=368±50, PH(18hs)=543±35*; Sh(24hs)=293±50; PH(24hs)=567±85*; % of oxidade glutathione (GSSG): Sh (3hs)=1.87±0.29, PH(3hs)=3.46±1.07; Sh(24hs)=2.75±0.20, PH(24hs)=10.87±0.60*. Superoxide Dismutase (SOD, expressed as % of Sh considered as 100% at all the times culture): Sh(1h, 3h, 18h, 24h)=100, PH(1h)=96.0±3.9; PH(3hs)=98.6±1.5; PH(18hs)=97.2±5.9; PH(24hs)=117.5±2.1*.
