LactobacillusplantarumCIDCA8327isanexopolysaccharide(EPS)-producerstrainisolatedfromkefirwithpromisingpropertiesforthedevelopmentoffunctionalfoods.Theaimofthepresentstudywastocharacter-izethestructureoftheEPSsynthesizedbythisstraingrowninskimmilkorsemidefinedmedium(SDM).Additionally,genesinvolvedinEPSsynthesisweredetectedbyPCR.L.plantarumproducesanEPSwithamolecularweightof104Dainbothmedia.WhengrowninSDMproduceanheteropolysaccharidecom-posedmainlyofglucose,glucosamineandrhamnosemeanwhiletheEPSproducedinmilkwascomposedexclusivelyofglucoseindicatingtheinfluenceofthesugarsource.FTIRspectraofthisEPSshowedsignalsattributabletoanα-glucan.Bothby1HNMRandmethylationanalysisitwaspossibletodeterminethatthispolysaccharideisabranchedα-(1→4)-D-glucancomposedof80%linearα-(1→4)-D-glucopyranosylunitsand19%(1→4)-D-glucopyranosylunitssubstitutedatO-3bysingleα-D-glucopyranosilresidues
