One of the fundamental steps in the diagnosis of diseases is the isolation and purification of antibodies from biological samples. Among different approaches, the use of magnetic nanoparticles (MNPs) functionalized with M protein (or A protein) linked to a Spy-tag has gained interest due to its efficiency and practicality. In this study, we rely on the Spy-tag/Spy-catcher chemistry to immobilize proteins on MNPs reducing the risk of protein denaturation that usually occurs in conventional immobilization processes, thus aiming to maintain the native conformation of the M protein. We synthesized magnetic nanoparticles through the co-precipitation method and subsequently functionalized them with the Spy-tag. To characterize these particles, we employed several techniques (VSM, TEM, IR, DLS, Z potential) and we obtained promising results indicating the successful binding between the Spy-tag and the MNP.These encouraging results motivate us to continue our investigation to assess the biological properties of these MNPs.
