Purification of biopharmaceuticals is generally more cost intensive than corresponding fermentation processes in the biotech industry, and can account for up to 80% of production costs. Affinity chromatography process using the Protein A ligand is one of the most important and expensive step, industrially used for monoclonal antibody production. In previous work our research group has prepared a novel chromatographic material based on poly(glycidyl methacrylate) grafted onto cellulose fibers. This material has been prepared by radiation-induced simultaneous grafting polymerization. Ion-exchange fibers prepared by this technique have shown high capacity at higher flow rates for ion-exchange chromatography of proteins. However, the immobilization of proteins have not been successfully performed.In this work are studied the preparation of an affinity chromatographic matrix containing a similar Protein A ligand (Avimer) based on radiation-induced grafted cellulose fibers.
