Caracterización Histopatológica de la Línea Tumoral CFM27 obtenida por pasajes singeneicos

Resumen:
Tumors obtained by induction in Wistar rats with n´nitroso n´methylurea (NMU), demonstrated an histopathological pattern correspondingto an alveolar mammary adenocarcinoma. Those originated inmice only treated with medroxiprogesterone (MPA) were ductalmammary carcinomas. Inductions with NMU in mice Balb/c didnot reproduce the tumorigenic effect. The induction and later promotionwith MPA in mice Balb/c generated an alveolar mammaryadenocarcinoma. After several syngeneic passages of tumoral fragmentsobtained by induction and promotion a tumoral line withdifferent histopathologic character was stablished. Mice of 5 weekswere induced with three doses of NMU (50 mg/kg) and three dosesof MPA (40 mg). Two months after induction tumors were developedand detectables by palpation. These were transplanted in syngeneicform to female mice without inmunosuppression. They werecarried out 29 successive passages in Vivo and samples were frozenat -120ºC to repeat passages starting from the same one. With alatency of 7 days they were developed tumors locally, differingfrom the original pattern to a sarcoma of small cells. With the histologicalroutine Tricrhomics were carried out and the cellular patternwas determined by inmunohistochemistry, using membranemarkers like Cadherin E, N, Cytoqueratin P8 and Pan cytoqueratine.The expression of receptor of hormon-dependence was evaluatedwith Progesterone monoclonals anti receptor of and anti proteinP29. Up to now the CFM-27 line maintains its histopathologic patternwithout changes in successive passages, being able to be atumoral model repetible for the study of different biological aspectsof neoplasia.
