Menopause is associated with higher prevalence of chronic diseases such as obesity and osteoporosis. Genistein (Gen) has been proposed as a natural therapy to counteract these diseases. Previously in our lab we demonstrated that, in the absence of ovarian function, obesity exhibited a deleterious action on bone metabolism through an enhancement in bone oxidative stress and reducing mineralization. In this work, we studied the effect of Gen on bone stress induced by obesity and hypoestrogenism. Females Wistar rats were bilaterally ovariectomized (OVX) and fed with a high-fat diet (OVX-Ob) for 10 weeks. Experimental systems: diaphysis of femurs (DF) and retroperitoneal AT explants isolated from OVX-Ob rats, or primary culture of DF-osteoblast (OBL). DF, AT, OBL were in vitro exposed to Gen for different interval of treatment.
