Executive summaryThe results presented in this second interim report correspond to the activities carried out from September 2022 to August 2023, under the agreement ?Obtención y caracterización de consorcios dehalogenantes autóctonos para ser utilizados en procesos de biorremediación de sitios contaminados con hidrocarburos?.The general objective of this agreement is to obtain and characterize dehalogenating bacterial consortia from samples of different impacted sites, to be used as inoculants in bioremediation processes of contaminated aquifers. This report shows the physicochemical and biological characterization of samples from Berazategui and other sites in Buenos Aires province. Physicochemical characterization included the determination of pH, humidity, conductivity at 25°C, total nitrogen, total phosphate, concentration of chlorinated hydrocarbons (tetrachloroethene, trichloroethene, 1,1-dichloroethene, 1,2 dichloroethene, vinyl chloride, carbon tetrachloride, chloroform, dichloromethane, 1,1,2,2 perchloroethane), concentration of trichlorofluoromethane, dichlorodifluoromethane and methanol, and detection of methane and ethene. For the biological characterization of the samples, DNA extraction was performed, followed by 16S rRNA gene metabarcoding analysis and quantification by TaqMan qPCR of 16S rRNA and tceA genes of Dehalococcoides. The set-up of the dechlorinating enrichment cultures is described in this report; 11 enrichment cultures that evidenced the capacity for a complete dechlorination of PCE to ethene and 7 enrichment cultures that produced DCM from CF were obtained.
