Resumen:
The search for more effective formulations to overcome biopharmaceutic drawbacks of drugs in the treatment of disease has led to the development and research of new drug nanocarriers that enable drug encapsulation and its release in a controlled spatiotemporal manner. Poloxamines, commercially known as Tetronic®, are amphiphilic block copolymers consisting of four arms of polyethylene oxide-polypropylene oxide linked to a central molecule of ethylenediamine. This structure confers the copolymers temperature- and pH-dependent self-aggregation properties. In this study, we preliminarily assessed the potential of two poloxamines and their conjugates with of a mannose derivative (a moiety that enables targeting to macrophages, the leishmania reservoir)to form polymeric micelles and encapsulate two FDA-approved drugs, nimodipine and amiodarone, that are currently investigated as alternative leishmanicidal agents in a second intended use; leishmaniasis is a neglected tropical disease.
