Decidualization is aprocess of endometrial transdifferentiation essential to sustainpregnancy. The events involved in decidualization are regulated bysteroid hormones progesterone and estradiol (E2) via the ligandactivated transcription factors progesterone receptor (PR) andestrogen receptor (ER). Endometrial specific transcription is alsoregulated by HOXs and FOXO transcription factors. In this work weused HiC to analyzed the changes in chromosome conformationorganization and RNA-seq to explore changes in gene expressionsignatures during in vitro decidualization of human t-HEC cell line.Decidualization was induced by exposure to cAMP, E2 and eitherprogesterone or the synthetic progestin R5020. In particular, wefocused on the Hox genes cluster of Chromosome 7 and on the humandecidualization signature, including among others Prl, IGFBP1 andLEFTY2. Preliminary results showed dramatic changes in loopingconformation in the HOX chromosome region and a correlation withchanges in decidual specific genes expression. Future analysis oftransposable elements enrichment and the distribution in chromatincompartments A and B (active and inactive) could provide a moredetailed explanation of the mechanisms that direct gene regulation toinduce decidualization in humans.
