Non-motor symptoms in a rat model of neurotoxicity induced by 6-OHDA: effect of intracisternal RAd IGF1 in anxiety – like behavior.

Parkinson's disease is a neurodegenerative disorder characterized by the progressive loss of dopaminergicneurons, resulting in a range of motor and non-motor symptoms. Although motor deficits are the main criteria fordiagnosis, there is growing evidence that non-motor symptoms may also play an important role in the diseaseprogression. In this study, we aimed to investigate the onset and progression of cognitive and anxiety-like behaviordeficits in a rat model of neurotoxicity induced by 6-OHDA infusion into the dorsolateral striatum. We assessed thebehavioral changes in male Wistar rats after 1, 2 and 3 weeks post-lesion using the Y-maze, elevated plus maze,light-dark box test and locomotor activity test. Our results showed anxiety-like behavior at 2 and 3 weekspost-lesion, before the onset of motor impairment. We also observed working memory impairment in 6-OHDA ratsafter 3 weeks of neurodegeneration, without motor alterations. Immunohistochemical analysis revealed a decreasein tyrosine hydroxylase immunoreactivity in the substantia nigra, ventral tegmental area, and striatum, indicatingpartial lesion of the nigrostriatal dopaminergic system.
