Resumen:
The Chinchilla lanigera is a species of high value livestock, The aim of this paper is to study the effect on some physiological parameters of different protectors used in the cryopreservation of epididymal sperm, in order to select the most appropriate for this technique, affecting as little as possible sperm quality of the samples. With sexually mature adult specimens recently slaughtered, obtaining the sperm by the tail epididymis puncture. We assessed: vitality, eosin test, membrane integrity by Hos test, fragmentation of DNA (acridine orange) and nuclear maturity (aniline blue). The acrosome reaction in capacitated sperm was induced with 20 µM progesterone.The cryoprotectants used (Test Yolk Buffer ® 20%, glycerol 6% + 62.5 mM sucrose, and 6% glycerol) were mixed with sperm suspension. The samples were centrifuged and pellets was freezing by vitrification with liquid nitrogen, Thawed at 37 ° C. The results indicate that the Test Yolk Buffer lesser degree affects motility and membrane integrity of spermatozoa in comparison with glycerol and sucrose. With regard to the vitality no significant differences were observed between cryoprotectant tested. Based on the above and the possible application in breeding, the cryoprotectant of choice is the Test Yolk Buffer that preserves sperm quality better.
