Vitamin D receptor polymorphisms in Individuals with Sporadic Cutaneous Malignant Melanoma

Introduction: Few studies have investigated the association between VDR polymorphisms and risk of melanoma however the results have been inconsistent. This study examined 38 VDR SNPs in individuals with single and multiple melanoma. Methods: DNA samples from 1208 individuals with multiple primary melanoma and 2470 with single primary melanoma recruited through an international multi-center population-based study were available for genotyping. All participants signed informed consent and completed an epidemiologic questionnaire. The genotyping included functional SNPs FokI, BsmI, 1A-1012 and 1A-1521 and tag SNPs. The laboratory methods included pyrosequencing, differential melting temperature (LightTyper), and MassSpectrometry (Sequenom). We investigated associations of VDR SNPs with risk to develop multiple melanoma using logistic regression. Results: Thirty-five of 38 SNPs were in Hardy-Weinberg Equilibrium. In analysis of VDR polymorphisms and risk for multiple melanoma 5/35 SNPs (rs10875712, rs4760674, rs11168287, rs2544028, rs2544038) modified the risk significantly by 24 to 37%, after adjusting for covariates (p
