The study of the tumor microenvironment is important to understand the biology of cancer. It has been known that the adipose tissue of breast cancer obese patient has higher TNF expression than the lean adipose tissue of breast cancer patient. Therefore, we have studied the effect of TNF on the lipid metabolism of murine mammary adipose tissue. In this model, TNF decreased adipocyte size and total lipid content, and increased glycerol secretion. With the aim of getting insights into the molecular pathways by which TNF is involved in the lipid metabolism of adipocytes, we used an in vitro model of 3T3-L1 cells differentiated into adipocytes. Hence, 3T3-L1 cells were stimulated with a differentiation cocktail and, after one week, the medium was changed for medium with or without TNF 20 ng/ml for 72 h.
