The effects of antioxidant or scavenger therapy on infarct size are still unclear. The aim of this study was to examine the efficacy of N-2-mercaptopropionyl glycine (MPG) on infarct size in a rat model. Isovolumic isolated rat hearts were subjected to 40 min coronary occlusion and 2 h reperfusion (R). Infarct size expressed as a percentage of risk area was determined by triphenyltetrazolium chloride (TTC) staining after 2 h of R. MPG 100 mM infusion during the first 20 min of R diminished infarct size (17.6±1.9 % vs 30.9 ±2.1% in untreated hearts). Postischemic contractility after MPG was also improved (after 2 h of R, left ventricular developed pressure was 71 ± 9% vs 51 ± 9 % in untreated hearts). These data show that MPG is cardioprotective and that free radicals produced early in reperfusion are involved in determining infarct size.
