Resumen:
The relationship between cellularsenescence and mitochondrial dynamic shift during thedevelopment of estrogen-induced pituitary tumours was recentlydescribed in our laboratory. Besides, it is known that energeticdeficiencies and cellular damage linked to the accumulation ofmitochondrial DNA mutations can be promoted by the estrogenoxidative action. In this context, we set out to evaluate in vitrothe possible 17ß-estradiol (E2) pro-oxidant actions onmitochondrial dynamics and the damage response activationunder pituitary tumour contexts. Pituitary tumour was induced inadult male Wistar rats by the subcutaneous implantation ofestradiol benzoate capsules (30 mg) for 10 days. Control group:implanted with empty capsules. Then, glands were collected anddispersed pituitary cells were cultured and exposed to E2 (1- 10-100 nM) for 15, 30 and 60 min. The ROS production wasanalyzed by flow cytometry and p-Nrf2 protein expression byimmunofluorescence.
