Chemical modification of proteins is of paramount interest in chemical biology, bioengineering,and biotechnology.[1?13] Owing to the great chemical diversity of proteins manifestedin the various functional groups found in amino acid side chains, the selectivemodification of proteins represents a considerable challenge. It is not only the chemicaldiversity, but also the delicate structure of proteins and enzymes that puts high demandson chemical reactions used for their modification. The potential of copper-catalyzedazide?alkyne cycloaddition (CuAAC) as an efficient method for selective protein functionalization has been investigated since shortly after its initial development.[14,15] Cowpea mosaic virus, decorated with an azide or alkyne, was the first natural protein investigated as a substrate in this regard.[16] Several important aspects of the CuAAC reaction for its successful use on proteins were recognized already in this study.
