The composition and function of animal gut microbiomes, key regulators of hostphysiology, are affected by temperature variation. Little is known about this inpoikilothermic vertebrates, whose physiology is also strongly influenced byenvironmental temperature. Here, we characterize the response of the gut microbiometo temperature in pacú (Piaractus mesopotamicus), an important aquaculture speciesnative to South America. Taking into account that there is increased evidence that gutmicrobiome disturbance may be an indirect way by which temperature impacts theanimal fitness, we aim to identify the microbial species present in the gut of Pacú fish,sensitive and tolerant to low temperatures. Toward this goal, we designed a highlymultiplexed protocol for 16S rRNA bacteriome profiling based on long read sequencingtechnology (Oxford Nanopore Technologies). To evaluate its taxonomic resolution, weused mock community samples as positive controls. Based on statistical analysis ofmicrobial diversity of the gut microbiome of P. mesopotamicus, it is concluded that lowtemperature (15ºC) affects the microbiota of sensitive individuals with significantchanges in their composition and structure, while the microbiota of tolerant individualsdid not experience significant changes. These results show that the thermal tolerance ofthe host has a significant effect on intestinal microbial diversity. Combined with futurestudies on the physiological response of the host, these results can inspire the design ofnew strategies for the cultivation of this species.
