Resumen:
Introduction. A composite of Hydroxyapatite (HAP)-containing poly-?Ã-caprolactone (PCL)-polydiisopropyl fumarate (PDIPF) (Blend) was developed as an alternative for bone tissue regeneration. We have previously investigated the physicochemical, mechanical and biocompatibility properties of the Blend using two osteoblast-like cell lines (UMR106 and MC3T3E1). They characteristic were compared with a blend without HAP and with a PCL/HAP film. The blend was shown to exhibit good compatibility and potential for use en bone regeneration. An important requirement for the use of these materials is to demonstrate low toxicity. This study aims to evaluate the toxicity of the films (PCL, PCL +1 % HAP, Blend and Blend + HAP) in a model of macrophage in culture. Materials and methods.  To evaluate the production of proinflammatory cytokines IL-1 and TNF (by ELISA) and NO (by the technique of Greiss), RAW 264.7 cells were cultured on membranes PCL, PCL +1 % HAP, Blend and Blend + HAP for 24, 48 and 72 hours. Cells grown on culture dish was used as control, while 0.1 ug / ml LPS was added as positive control. Result. There weren?Lt significative diference in production of NO and cytokines for RAW 264.7 cells grown on films and culture dish. Conclusion. The films shown to have low toxic effect showing that they can be used in the area of bone tissue regeneration.
