Resumen:
7sage of glyphosatebased herbicides )$Hshas been expandedover the lastdecade. 4ecently, Ye found that oral exposureto a )$H in rats during perinatal period pregnancy andlactation produced subfertility as a consequence of implantation failures. +mplantation is a complex process regulated by endocrinesignaling pathYays primary mediated via steroid receptors, such as estrogen receptor alpha E4a. +t has been identifiedfive promoters called E, 1T, 1, 10, and 1S Yhich control E4atranscription initiation yielding differenttranscripts Yith alternativey untranslated regions. The purpose of this YorMYas toinvestigate the effects of in utero and lactational exposure to a)$H over the control of E4a expression in the preimplantationuterus. 2regnant rats F Yere orally exposed to mg )$H/-g/day through food from gestational day )& until Yeaningpostnatal day , 20&. 9hen F females reached the sexualmaturity 20& Yere mated Yith males of proven fertility, andpregnancy Yas confirmed by vaginal smears. 2regnant F femalesYere sacrificed on )& preimplantation period, and uterinesamples Yere collected and store at uC until analysis. 4elativeexpression of total E4a m40A and E4a transcripts containingalternative ŏuntranslated regions E, 1T, 1, 10, and 1S Yasevaluated by real time 4T2C4. First Ye found that during thepreimplantation period, E4a gene transcription Yas regulated by1S, 1, 1T and E promoters, regardless of treatment group. 9henYe analyzed )$H effects, Ye detected an increase of total E4am40A expression. /oreover, the increase in total E4a m40A in)$H group Yas mediated by an increased expression of E4a1variant. These findings shoYed that perinatal exposure to a )$Halters E4a expression during the preimplantation period, by affectingtherelative abundance of alternative ŏuntranslated regionof E4a transcripts in the uterus. These alterations could explain,at least in part, the )$Hmediated subfertility.
