Resumen:
)lyphosate exposure during critical periods of developmentinduced adverse effects on the reproductive system of male rats, suggesting an endocrine disruption action. In addition, it has beenreported that the male rat mammary gland /) is affected bythe action of endocrine disruptors. Here, Ye evaluated Yhetherperinatal exposure to a glyphosate based herbicide )$H modifiesthe /) development of pre and postpubertal male animals.2regnant rats F Yere exposed orally through the diet to vehicleC, saline solution or mg/Mg/day of )$H from gestationalday until Yeaning. 1n postnatal day 20& and 20&F males Yere sacrificed and /) and blood samples Yere collected. The /) samples Yere processed for Yhole mount 9/or Mept at uC for 4T2C4 analysis. 1n 20& and 20&serum levels of testosterone T Yere measured and 9/s Yereanalyzed. 1n 20&, the m40A expression of estrogen receptoralpha ES4 and androgen receptor A4 Yas also evaluated. The treatment Yith )$H produced no signs of abnormalmaternalor nursing behaviors, or changes in the body Yeight gain orpellet consumption of the F dams. The length of gestation Yasunaltered, and no gross malformations Yere observed in the Fpups at delivery or Yeaning. 0o differences Yere observed in Tserum levels betYeen experimental groups and T concentrationYas higher on 20& C .v. ng/ml )$H .v.ng/ml compared to 20& C .v. )$H .v. ng/ml. 1n 20&, the treatment Yith )$H reduced /) total area.+n contrast, on 20&, total area, longitudinal groYth and perimeterYere increased in )$Hexposed group no differencesYere observed in the epithelial area. +n addition, )$H treatmentdecreased the m40A expression of ES4 on 20& animals,Yhereas A4 m40A levels Yere increased. 1ur results demonstratethat perinatal exposure to )$H alters /) morphology andgene expression long after exposure ended and suggest that theeffects may be due to endocrine disruption.
