NITRATION OF PLACENTA AQUAPORINS AFFECTS THE SURVIVAL OF VILLOUS TROPHOBLAST CELLS

Objectives: Our aim was to evaluate nitrative stress effect on placentalAQPs and its role in the pathogenesis of preeclampsia.Methods: This study was approved by the ethics committee of Hospital Nacional Prof. Dr. A. Posadas. To search putative sites of nitration, the amino acid sequences of AQP3, AQP4, AQP8 and AQP9 were analyzed by in silico analysis. Levels of each 3NTp-AQP were evaluated by immunoprecipitation in preeclamptic and normal placentas. To study the effect of nitration on AQPs expression and function, explants from normal placentas were culture in the presence of ONOO- (100mM) to induce nitrative stress. Damage cells indexes, cell viability and cytotoxicity were determined. Water uptake and AQP molecular expressions were also studied. The survival of trophoblast cells was evaluated in low glucose medium with or without L-lactate and before and after the inhibition of AQPs.Results: In silico analysis showed that AQP9 has more probability of beingnitrated than the others AQPs studied.
