Objectives: Magnesium sulfate (MgSO4) is the drug of choice to prevent seizures in preeclamptic pregnant women, but its mechanism of action at the molecular level still is an enigma. We aimed to evaluate the effect of MgSO4 on AQP9 expression and function in human trophoblasts exposed to nitrative stress.Methods: This study was approved by the ethics committee of the Hospital Nacional Dr. Prof. A. Posadas. Placental explants were cultured in complete DMEM-F12 in the presence and absence of 200 mM peroxynitrite (ONOO-) to induce oxidative and nitrative stress. 4 mM MgSO4 was added to neutralize the effect of ONOO-. Lipid peroxidation (TBARS), superoxide dismutase 1 (SOD-1), and catalase expressions were analyzed.
