Resumen:
Secretory leukocyte protease inhibitor(SLPI) is a serine proteaseinhibitor with pleitropic activities on immune andnon-immune cells.Lately, it has been proposed SLPI as a biomarker of acutekidneyinjury with a protective role on tubular epithelial cells. The aim ofthepresent work was to unravel whether SLPI action on kidney is animmunemediated effect. We found that the administration of SLPIreduced the kidneydamage in a rat model of ischemia reperfusioninjury. However, SLPI was alsoable to reduce the damage in a gen-tamicin-induced nephrotoxicity rat model. Inin vitro experiments, theHK-2 human epithelial cells-treatment with SLPIreduced the cellapoptosis induced by serum starving conditions or calcineurinin-hibitors. A real-time PCR array data with HK-2 cells revealed thatSLPImodulated transcript levels of genes mainly involved in DNArepair, response tomisfolded proteins and inflammatory responsepathways. Furthermore, SLPI favorsthe proliferation and migrationof HK-2 cells. In kidney transplant patients,plasma levels of SLPIwere high and indirectly associated with kidney functionbut alsowith leukocyte proliferation index. However, the levels of SLPItran-scripts found in the renal biopsies were indirectly associated withplasmacreatinine and TGFb but directly associated with HMOX-1.An indirect associationwas found between plasma levels of FK506and SLPI. These results confirm thatlocal SLPI exert a protectiverole in kidney transplant patients which can bemediated directlyon epithelial cells and indirectly by affecting the immuneresponse.Furthermore, it suggests that immunosuppression treatment can re-ducethe expression of this nephropotective factor
