Asociación de patrones seroindeterminados por Western Blot con bajos niveles de carga proviral en individuos infectados con el Virus Linfotrópico T Humano tipo 1

Resumen:
According to HTLV-1/2 diagnosis algorithm, reactive screening results are generally confirmed by Western Blot (WB). However, the large number of indeterminate WB patterns is a frequent problem worldwide. The aim of this study was to determine whether low levels of proviral load (PVL) are associated with seroindeterminate results by WB. Therefore, PVL was determined in 49 samples, classified as positive by WB from individuals with disease (G1; n= 27) of which 5 corresponded to Leukemia/Lymphoma patients and 22 to HTLV-1 Associated Paraparesis (HAM) ones, samples positive by WB from asymptomatic carriers (G2; n = 18) and seroindeterminate samples by WB (G3; n= 4). All samples were confirmed by n-PCR. The viral gen pol and albumin were quantified by real-time SYBR Green PCR (ABI Prism System- AppliedBiosystems 7500). PVL values were expressed as HTLV-1 copies/100 PBMCs. Calibration curves were constructed using a DNA stock from MT2cell line (Limit of quantification: 3 pol copies / reaction - R2> 0.99) and the analysis was performed by Krustall Wallis (InfoStat v.2011).
