The impact of aerobic training in cardiovascular alterations and cognitive processes following ovariectomy: the key role of IGF1

During menopause, women are exposed to an increase in cardiovascular risk (CVR), associated with lack of estrogens. Sodium/proton exchanger (NHE) and Sodium/bicarbonate cotransporter (NBC) are regulators of the cardiomyocyte pH. NHE hyperactivity and the reduced activity of the electrogenic isoform of NBC (NBCe1) are correlated with increased CVR. Estrogen deficiency is associated with cognitive changes. Aerobic exercise exerts beneficial effects on cardiovascular health and cognitive processes.Bilateral ovariectomy was performed in young female Wistar rats (OVX), sham operated animals (Sham) were used as control. OVX rats were randomly assigned to a sedentary group (OVXs) or to an aerobic swimming routine (8 weeks/5 days a week) (OVXe). Another group of OVX rats were intramuscularly injected with a recombinant adenoviral construct harboring the cDNA of human Insulin like growth factor 1 (IGF1). OVXs cardiomyocytes were cultured and treated with IGF1 (1uM) to measure the impact in ROS production.Aerobic training induces physiological hypertrophy in OVXe. NHE activity was increased in OVXs, and both exercise and IGF1 abolished this alteration (JH mmol/min: Sham 1.42±0.24; OVXs 2.15±0.23*; OVXe 1.22±0.24; OVXIGF1 0.82±0.15) (JH mmoles/min: Sham 1.42±0.24 OVXs 2.15±0.23*; OVXe 1.22±0.24; OVXIGF1 0.82±0.15). NBCe1 activity was decreased in OVXs, and exercise prevented it (DpH: Sham 0.25±0.02; OVXs 0.09±0.02*; OVXe 0.14±0.01) (pH Sham 0,25±0,02 OVXs 0,09±0,22*; OVXe 0,15±0,1). Antioxidant capacity was impaired in OVXs, and recovered after the training. OVXIGF1 rats showed an improvement in long-term memory and exploratory behavior, both of which were affected by ovariectomy. IGF1 decreased ROS production in OVXs cultured cardiomyocytes.Our results show that aerobic training has a positive impact in the remodeling of alkalinizing mechanisms observed in OVXs, as well as an increase in antioxidant capacity, and this is, at least partially, due to IGF1 signaling. Our data also support the involvement of sex hormones in cognitive functions in female rats and suggest a positive effect of transgenic IGF1.
