Resumen:
Acetylcholine (ACh) is the major neurotransmitter of Parasympathetic Nervous System. The functions of ACh are mediated through muscarinic (mAChR) and nicotinic receptors (nAChR). The ACh plays a key role in allergic diseases, as well as the Thymic Stromal Lymphopoietin (TSLP), which triggers dendritic cell?mediated T helper (Th)2 inflammatory responses. Previously, we describe the presence of mAChR, ChAT and AChE (enzymes that synthesized and hydrolyzed ACh, respectively) in human myeloid dendritic cells (DC) and its activation by cholinergic agonists. In this work, we evaluated the TSLP-activated DC (TSLP-DC) physiological modulation by ACh. The monocytes were isolated from peripheral blood obtained from healthy nonsmoker donors, by positive selection (CD14, Miltenyi, % purity >95), and then were cultured with GM-CSF and IL-4 for 5 days for obtain DC.
