Resumen:
Background Dendritic cells (DC) and monocytes (Mo) express a non-neuronal cholinergic system although its role in early pregnancy is unknown. We evaluated the effects of endogenous acetylcholine (ACh) and VIP on trophoblast cells (Tb), DC/Mo migration to Tb and Tb- DC/Mo interaction. Methods Mo purified from blood mononuclear cells of healthy women by MACS were differentiated or not to DC. Expression of choline-acetyltranferase (ChAT), acetylcholinesterase (AChE) and muscarinic receptors (mAChR) was evaluated in Tb line Swan-71 by RTPCR and Western blot; cell migration to Tb by FACs and MCP-1, IL-10 and TNF-a by ELISA in cocultures with Neostigmine (Neo) or VIP. (Protocol approved: Academy of Medicine Ethics Committee) Results Tb express ChAT, AChE and m1-m5/AChRs. Neo and VIP enhanced chemokine production (que quimoquinas MCP-1 y no se si medistes algo mas) in Tb and its conditioned medium induced DC and Mo migration. Tb-DC interaction raised IL-10 and Neo-induced MCP-1. Conclusion Tb express a complete cholinergic system that modulate Tb-DC interaction and Mo migration.
