Synergic activity of oligostyrylbenzenes with amphotericin B against Candida tropicalis biofilms

Resumen:
Antimicrobialdrug resistance is a serious challenge inclinical settings worldwide, withbiofilm formation having been associated withthis problem. In the presentstudy, the synergism of oligo(styryl)benzene (OSBs)compounds in combinationwith amphotericin B (AmB) against Candidatropicalis biofilmswas investigated. In addition, the toxicity inhuman blood cells wasdetermined. Synergistic combinations of OSBs and AmB wereevaluated to considerfuture effects of OSBs in vivo. The checkerboardmicrodilution method was used tostudy the interactions of one anionic (1)andtwo cationic (2 and 3) OSBs with AmB. We investigated theeffectsof OSBs on reactive oxygen species (ROS) and the levels of the reactivenitrogenintermediates (RNI). The cellular stress affected biofilm growththrough anaccumulation of ROS and RNI, at synergistic concentrations of OSBsand AmB.Furthermore, significant surface topography differences were notedupontreatment with the OSB2/AmB combination, using confocal laserscanningmicroscopy in conjunction with the image analysis software COMSTAT. The resultsrevealeda low toxicity of leukocytes and red blood cells at synergisticcombinations ofcationic OSBs with AmB. Thesefindings demonstrated theantibiofilm effects of OSBs and the synergism of AmBwith cationic OSBs againstbiofilms of C. tropicalis for the first time.
