The mechanism by which estradiol (E2) regulates adiposetissue (AT) development are largely unknown. Previously, wehave shown an in vitro E2-dependent decreased adipogenic potentialin retroperitoneal adipose tissue (RPAT) adipocyte precursorcells. In this study, our aim was to assess the role of E2 deficiencyon RPAT and inguinal AT (IAT) pads in female rats. S-D rats were dividedinto three groups: non-ovariectomized (Sham operated, CTR),ovariectomized (OVX) and ovariectomized pair-fed (OVX PF), latterones castrated prepubertally. CTR and OVX food consumption wasad libitum, while for OVX PF was the same to CTR, due to a higherfood consumption of OVX rats. Food intake and body weightswere recorded daily from surgery (27-day-old) to experimental day(60-day-old). At this time, trunk blood was collected to measure plasmametabolites. RPAT and IAT pads were dissected, weighed andhistological analysis were performed. Moreover, stromal vascularfraction (SVF) from both tissues was isolated to test mRNA levelsof PPARγ (adipogenic marker) by qPCR. OVX PF animals showa decrease in RPAT mass and %RPAT (OVX PF P
